Candida steals arachidonic acid from host cell membranes and by using its own internal cyclooxygenase (COX)-like enzyme pathways, it converts this stolen arachidonic acid into PGE2. PGE2 acts as a localized fungal hormone (*) that stimulates germ tube formation (the very first step of hyphal growth). So without PGE2, Candida struggles to form these hyphal meshes. (*) PGE2 enhances the expression of specific fungal surface proteins called adhesins (such as HWP1 and ALS3). Then the vicious cycle starts were mature Candida biofilms secrete significantly more PGE2 than free-floating yeast cells and this massive surge of extracellular PGE2 signals neighboring cells to accelerate the production of the extracellular polymeric substance (EPS) matrix. The high concentrations of PGE2 within the biofilm matrix correlate directly with upregulated efflux pumps. This allows the mature biofilm to aggressively pump out antifungal medications, making it up to 1,000 times more drug-resistant than single Candida cells In case you are not aware, biofilms are organized “groups” of microorganisms such as bacteria, fungi, archaea, or protozoa commonly formed in the gut, oral cavity and sinuses. They are embedded in a self-produced extracellular matrix composed of polysaccharides, proteins, and extracellular DNA (eDNA) that acts as a physical barrier and makes them highly resistant to antibiotics and antifungals. Biofilm formation starts with planktonic microbes adhering to a surface through something called van der Waals forces or specific adhesins, then microbes start producing matrix components.
